Parenteral Administration","legacy_section_text":"<ol style=\"list-style-type: lower-alpha\" start=\"1\">\r\n \t<li>Substances administered <a class=\"gtip\" href=\"\/animalcare\/glossary\/parenteral\">parenterally<\/a> should be:\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li>Isotonic (the same concentration of solute as the blood)<\/li>\r\n \t<li>Close to physiologic pH (6.8-7.2)\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>If pH is outside of physiologic range, administer the substance through a central vessel (such as the jugular or femoral vein) or buffer the solution such that pH is appropriate.<\/li>\r\n<\/ol>\r\n<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>All substances given parenterally\u00a0<strong>must be sterile<\/strong>\u00a0and should be delivered aseptically.<\/li>\r\n \t<li>If the preparation is not a commercially manufactured solution, it must be mixed in a laminar flow hood or <a class=\"gtip\" href=\"\/animalcare\/glossary\/biological-safety-cabinet-bsc\">biosafety cabinet<\/a> and filtered through a 0.2 micron filter.<\/li>\r\n<\/ol>","legacy_media_position":"","legacy_media_file":"","legacy_media_url":"","legacy_glossary_term":"","legacy_glossary_nids":"","legacy_resource":"","legacy_resource_nids":"","legacy_buttons":null},{"legacy_section_type":"text_area_with_subheading","legacy_heading":"","legacy_subheading":"3. Routes of Parenteral Administration","legacy_section_text":"<ol style=\"list-style-type: lower-alpha\" start=\"1\">\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/iv\">Intravenous<\/a>\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li>Substances can be administered as a bolus or as an <a class=\"gtip\" href=\"\/animalcare\/glossary\/infusion\">infusion<\/a> (given over &gt; 5 minutes).\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>The maximum <a class=\"gtip\" href=\"\/animalcare\/glossary\/bolus\">bolus<\/a> injection volume is 1 ml\/kg. If larger quantities are administered, give as an infusion.<\/li>\r\n \t<li>Infusions are often administered with specific equipment (precision pumps or microdrip infusion sets).<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>Site selection for venous access is species-specific. Common sites are listed below:\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Rodents: Lateral tail vein, saphenous vein, or retro-orbital venous sinus<\/li>\r\n \t<li>Rabbits: Lateral ear, jugular, or cephalic vein<\/li>\r\n \t<li>Larger species: Jugular, cephalic, femoral, or saphenous vein<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>Consult with veterinary staff for recommendations on refinements to improve animal comfort during repeated IV dosing.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/intraperitoneal-ip\">Intraperitoneal<\/a>\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li>Injections are administered into lower abdominal quadrants. Aspirate before injecting to avoid inadvertent administration into the bladder or gastrointestinal tract.<\/li>\r\n \t<li>Repeated daily intraperitoneal dosing for up to one month is well-tolerated in rodents. Doses should be administered to alternating sides of the abdomen.<\/li>\r\n \t<li>Administration of irritating substances may cause ileus (stasis of the gastrointestinal tract) and peritonitis (inflammation of the abdominal cavity).<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>Administration to the skin or muscle <em>(see\u00a0<strong>Appendix A<\/strong>)<\/em>\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/topical-epicutaneous\">Topical<\/a> (epicutaneous)\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Avoid application of caustic or irritating substances.<\/li>\r\n \t<li>Apply substances to skin that is unbroken and free of hair.<\/li>\r\n \t<li>Avoid application of substances to sites available for grooming by the animal.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/transdermal-percutaneous\">Transdermal<\/a> (percutaneous)\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Transdermal dosing is typically accomplished by application of a patch impregnated with the substance of interest.<\/li>\r\n \t<li>Apply the patch so as to avoid inadvertent ingestion or removal by the animal.<\/li>\r\n \t<li>Systemic absorption is not immediate. Patches should be applied prior to the time of anticipated need according to manufacturer's instructions.<\/li>\r\n \t<li><strong>Do not cut patches<\/strong>\u00a0to reduce dose size. If an appropriate dose of patch is not commercially available, consider an alternative route of administration.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/intradermal-id\">Intradermal<\/a>\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Use a small, sharp needle (25-27G).<\/li>\r\n \t<li>Tent the skin. Holding the syringe parallel to the animal, direct the needle into the dermis. Aspirate and inject.<\/li>\r\n \t<li>Inadvertent subcutaneous administration is common. Consult ULAM veterinary or training core staff for assistance.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/sc\">Subcutaneous<\/a>\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Tent the skin. Holding the syringe parallel to the animal, direct the needle into the subcutis. Aspirate and inject.<\/li>\r\n \t<li>The rate of absorption from the subcutis may be slower than with other parenteral routes.<\/li>\r\n \t<li>Subcutaneous infusions can be administered with the use of an oily depot or osmotic minipump. Consult veterinary staff for additional information.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/im\">Intramuscular<\/a>\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>IM dosing is best used in larger species with greater muscle mass.<\/li>\r\n \t<li>In smaller animals, use the gluteal or quadriceps muscles.<\/li>\r\n \t<li>In larger animals, use the gluteal, quadriceps, biceps or epaxial muscles.<\/li>\r\n \t<li>Take care to avoid the sciatic nerve which runs along the caudal aspect of the femur. Inadvertent injection into nerves can result in paralysis and localized muscle necrosis.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/intracranial\">Intracranial<\/a>\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Intracranial injections require <a class=\"gtip\" href=\"\/animalcare\/glossary\/anesthesia\">anesthesia<\/a> and stereotactic equipment. Injections can be administered through a surgically implanted cerebral cannula, direct injection, or an osmotic pump catheter.<\/li>\r\n \t<li>Animals\u00a0<strong>must<\/strong>\u00a0be heavily sedated or anesthetized for cannula or catheter placement and for direct injections<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>Epidural or <a class=\"gtip\" href=\"\/animalcare\/glossary\/intrathecal-it\">intrathecal<\/a><em> (see\u00a0<strong>Appendix B<\/strong>)<\/em>\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li>Epidural or intrathecal administration of substances requires highly trained personnel. Consult with veterinary staff before attempting this technique.<\/li>\r\n \t<li>Animals\u00a0<strong>must<\/strong>\u00a0be heavily sedated or anesthetized.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>Inhalational administration\r\n<ol style=\"list-style-type: decimal\" start=\"1\">\r\n \t<li><a class=\"gtip\" href=\"https:\/\/az.research.umich.edu\/animalcare\/glossary\/intranasal-in\/\">Intranasal<\/a>: Use the smallest possible volume to avoid suffocation. Systemic absorption is rapid.<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/intratracheal-it\">Intratracheal<\/a>: This technique requires intubation or surgical access to the trachea. Enteral Administration","legacy_section_text":"<ol style=\"list-style-type: lower-alpha\" start=\"1\">\r\n \t<li>Voluntary consumption\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li>Substances are typically mixed with the daily diet, flavored water, or other palatable items to encourage consumption. Care should be taken to maintain an appropriate daily caloric intake and to habituate animals to any novel food items before adding drug.<\/li>\r\n \t<li>Care should be taken to ensure animals consume all agent offered. Laboratory personnel are responsible for ensuring that food and water intake is adequate.<\/li>\r\n \t<li>Food or water containing additives should be clearly labeled and disposed of properly.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li>Gavage\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/gavage\">Gavage<\/a> is often used to administer an exact <a class=\"gtip\" href=\"https:\/\/az.research.umich.edu\/animalcare\/glossary\/per-os-po\/\">PO<\/a> dose.<\/li>\r\n \t<li>Administration of gavage volumes greater than 5 ml\/kg may cause distress in species that are unable to vomit such as mice.<\/li>\r\n \t<li>The gavage tube size should be appropriate for the species being dosed. Contact veterinary staff for assistance.<\/li>\r\n<\/ol>\r\n<\/li>\r\n \t<li><a class=\"gtip\" href=\"\/animalcare\/glossary\/rectal\">Rectal<\/a>\r\n<ol style=\"list-style-type: lower-roman\" start=\"1\">\r\n \t<li>This technique is not frequently used in laboratory animals.
